I have been reading the comic from near the beginning, enjoy it, never read the fanfic novel its is based on?

Anyway, I have a question, hope this is the right place.

So, Ehlissa is an original character, right? The other characters are based on the old school, Gygax Greyhawk campaign characters, but Ehlissa is an addition? I assume she is inspired in some way by Ehlissa of Nightingale fame?

I believe she's based on Gary Gygax's daughter Elise Gygax's PC. The recollections by Gary Gygax and Rob Kuntz that I've read didn't give her character a name, but she was part of the original Castle Greyhawk adventure and Ehlissa is named after Elise Gygax. No connection to Queen Ehlissa of the Nightingale except a common namesake.
